How does Deutivacaftor + tezacaftor + Vanzacaftor work?
Vanzacaftor and tezacaftor help selected abnormal CFTR proteins reach the cell surface. Deutivacaftor helps the CFTR channel open more effectively, improving chloride and water movement and reducing abnormally thick secretions.

Strengths: 50 mg + 20 mg + 4 mg, 125 mg + 50 mg + 10 mg
How to take Deutivacaftor + tezacaftor + Vanzacaftor
Follow the specialist CF team’s instructions.
Take the tablets with a meal or snack containing fat.
Swallow tablets whole; do not chew, crush or break them.
Avoid grapefruit-containing food and drink.
Continue other prescribed CF treatments unless your clinician changes them.

- Headache

Deutivacaftor + tezacaftor + Vanzacaftor in pregnancy
Pregnancy experience is limited. Continuing or starting treatment should be agreed with the specialist CF and maternity teams after weighing maternal benefit against uncertainty. Do not stop treatment without advice.
Deutivacaftor + tezacaftor + Vanzacaftor while breastfeeding
It is unknown whether this combination enters human milk, and risk to a breastfed infant cannot be excluded. The specialist team should weigh the benefits of breastfeeding and maintaining effective maternal CF treatment.
Deutivacaftor + tezacaftor + Vanzacaftor interactions
Check all prescribed, non-prescribed and herbal products with the CF team because important interactions can occur.
Rifampicin, rifabutin, carbamazepine, phenytoin, phenobarbital, efavirenz or St John’s wort
May substantially reduce treatment exposure and effectiveness; combined use is not recommended.
Azole antifungals, clarithromycin, erythromycin, telithromycin or verapamil
May increase exposure and require specialist review.
Grapefruit
May increase exposure and should be avoided.
