How does tacrolimus inhibit T-cell activation at the molecular level?

Tacrolimus appears to bind cytosolic FKBP12, forming a complex that specifically and competitively binds to and inhibits calcineurin, causing calcium-dependent inhibition of T-cell signal transduction and preventing transcription of a discrete set of lymphokine genes.
