Itraconazole increases ivacaftor exposure 4.3-fold without affecting lumacaftor exposure in Orkambi; no dose adjustment is necessary when itraconazole is started during established Orkambi treatment, but Orkambi dosing should be adjusted when it is started in patients already taking itraconazole.
