How does Sebetralstat work?
Sebetralstat reversibly blocks plasma kallikrein. This reduces formation of bradykinin, which increases blood-vessel leakage and causes HAE swelling.

Sebetralstat in pregnancy

Strong CYP3A4 inhibitors, including itraconazole, ketoconazole, ritonavir or clarithromycin
May substantially increase sebetralstat exposure, requiring review of the attack plan.
Strong or moderate CYP3A4 inducers, including phenytoin, carbamazepine, phenobarbital, rifampicin or efavirenz
May markedly reduce sebetralstat exposure and potentially reduce its effect.
Metformin
Sebetralstat may increase metformin exposure through renal transporter inhibition. Consider renal function and lactate monitoring in people at higher risk of lactic acidosis.
