Treatment of adults with relapsed and/or refractory multiple myeloma after at least two previous regimens that included bortezomib and an immunomodulatory agent.
Used with bortezomib and dexamethasone, not on its own.
How does panobinostat lactate anhydrous work?
Panobinostat blocks histone deacetylase enzymes. This alters gene activity and can promote growth arrest or death in susceptible myeloma cells.

How to take panobinostat lactate anhydrous
Follow the specialist treatment calendar exactly; take panobinostat only on its scheduled days.
Swallow capsules whole with water at the same time of day.
Take with or without food.
Do not open, crush or chew capsules.
Avoid grapefruit, grapefruit juice, star fruit, pomegranate and pomegranate juice.
If you vomit after taking a capsule, do not take a replacement; follow your treatment calendar for the next capsule.

Common or expected effects
- Diarrhoea, nausea, vomiting, abdominal discomfort and reduced appetite
- Tiredness, weakness, dizziness, headache or difficulty sleeping
- Low platelets, red cells, white cells or neutrophils
- Infections, including respiratory infections and pneumonia
- Low potassium, phosphate or sodium levels
- Low blood pressure, breathlessness, cough, fever or peripheral swelling
- Weight loss
Get urgent medical advice
- Fever, chills, confusion or worsening breathlessness, which may indicate serious infection or sepsis
- Unusual bruising, blood in vomit or stools, coughing blood or other significant bleeding
- Severe or persistent diarrhoea or vomiting, reduced urination, dizziness or other signs of dehydration
- Fainting, chest pain, sustained palpitations or an irregular heartbeat
- Severe weakness or breathlessness associated with markedly reduced blood-cell counts

Panobinostat may seriously harm an unborn baby. Pregnancy testing is required before treatment for people who could become pregnant, and highly effective contraception is needed during treatment and for the product-specified period afterwards. Hormonal contraception should be supplemented with a barrier method. Contact the oncology team promptly if pregnancy occurs or is planned.

Do not breastfeed during treatment. Transfer into milk is unknown and serious toxicity to an infant is possible. Discuss feeding plans with the oncology and maternity teams.

Strong CYP3A or P-glycoprotein inhibitors, including azole antifungals, ritonavir and saquinavir
These can increase panobinostat exposure and toxicity.
Strong CYP3A inducers, including carbamazepine, phenytoin, phenobarbital, rifampicin, rifabutin and St John’s wort
These may lower panobinostat exposure and reduce effectiveness; concomitant use should be avoided.
Medicines that prolong the QT interval
Combined use may increase the risk of a dangerous heart-rhythm disturbance.
