How does erlotinib inhibit EGFR signalling?

Erlotinib inhibits EGFR tyrosine kinase and intracellular phosphorylation; in EGFR mutation-positive tumours, tight binding to the ATP-binding site in the mutated kinase domain blocks downstream signalling, stopping cell proliferation and inducing death through the intrinsic apoptotic pathway.
