Amelogenin is encoded by the AMELX gene on the X chromosome (and a smaller copy, AMELY, on the Y chromosome). Mutations in AMELX cause X-linked amelogenesis imperfecta. Amelogenin comprises approximately 90% of the enamel matrix protein during development and is progressively removed during enamel maturation, leaving a highly mineralised (96% inorganic) mature enamel.
