What is Acoramidis hydrochloride used for?
Acoramidis is licensed for:
Wild-type or variant transthyretin amyloidosis in adults with cardiomyopathy (ATTR-CM).
How does Acoramidis hydrochloride work?
In ATTR-CM, the four-part transthyretin protein can separate into unstable components that misfold and form amyloid deposits in the heart. Acoramidis binds to and stabilises transthyretin, reducing this separation and slowing amyloid formation.

- Gout, causing painful joint inflammation.

- Severe allergic reaction, including sudden facial, mouth or throat swelling, breathing difficulty, faintness or collapse.
Acoramidis hydrochloride in pregnancy
There are no data from pregnant women, and animal studies found developmental toxicity alongside maternal toxicity. Acoramidis is not recommended during pregnancy or for women who could become pregnant and are not using contraception. Discuss pregnancy planning or suspected pregnancy promptly with the prescriber.
Acoramidis hydrochloride while breastfeeding
It is unknown whether acoramidis or its metabolites enter human milk, and a risk to the infant cannot be excluded. The UK product information states that it should not be used during breastfeeding.

CYP2C8 or CYP2C9 substrates with a narrow therapeutic index

A dedicated interaction study has not been performed, although the main clinical study found no difference in acoramidis exposure or TTR stabilisation.
Diuretics
